Emerging fungal infection in South West U.S. mimics cancer
ScienceDaily (Mar. 29, 2012) ? An emerging fungal infection of the gastrointestinal tract that mimics cancer and inflammatory bowel disease appears to be emerging in the Southwestern United States and other desert regions, according to Mayo Clinic researchers in Arizona investigating the disease. The invasive fungus, Basidiobolus ranarum, is typically found in the soil, decaying organic matter and the gastrointestinal tracts of fish, reptiles, amphibians, and bats.
Mayo researchers studied 44 cases of human gastrointestinal basidiobolomycosis reported from around the world, including 17 from Arizona, one from southern Utah and one from elsewhere in the U.S. Eight of the 44 patients died. Mayo's review of the cases is published online in the journal Clinical Infectious Diseases.
Basidiobolomycosis is usually a subcutaneous infection in the tropical and subtropical regions of the world that develops following traumatic inoculation of the fungus under the skin. The emergence of gastrointestinal involvement with Basidiobolus in arid regions has been considered unusual.
"The exact mode of acquisition of this gastrointestinal infection is unclear, although consumption of contaminated food or dirt is the favored hypothesis," says lead author H.R. Vikram, M.D., an infectious diseases physician at Mayo Clinic, where seven of the 19 U.S. cases studied were treated. "The infection is still considered so rare that no one had put together a complete description." He adds that more study needs to be done to determine how this infection is contracted, what underlying diseases might predispose patients to this infection and how best to treat it. He emphasizes that early recognition is key to successful treatment.
The first U.S. case of gastrointestinal Basidiobolus infection was reported in 1986. The CDC subsequently investigated six cases in Arizona between 1994 and 1999. This sparked the interest of researchers at Mayo Clinic in Arizona to study this infection.
Patients with this rare fungal infection had non-specific symptoms such as abdominal pain or a mass that could be felt on examination. Before a conclusive diagnosis of the fungal infection was made, most patients were thought to have an abdominal cancer, inflammatory bowel disease or diverticulitis. Surgical resection of the area of involvement and prolonged antifungal therapy successfully treated most patients.

Children who develop asthma have lung function deficits as neonates, study suggests
ScienceDaily (Mar. 30, 2012) ? Children who develop asthma by age seven have deficits in lung function and increased bronchial responsiveness as neonates, a new study from researchers in Denmark suggests.
"Previous research on the relationship between neonatal lung function and the development of asthma has been conflicting," said lead author Hans Bisgaard, MD, DMSci, professor of pediatrics at the University of Copenhagen and head of the Danish Pediatric Asthma Centre. "Our study shows that children with asthma by age seven already had significant airflow deficits and increased bronchial responsiveness as neonates. Lung function deficits also progressed throughout childhood in our study, suggesting a potential opportunity for early intervention."
The findings were published online ahead of print publication in the American Thoracic Society's American Journal of Respiratory and Critical Care Medicine.
The prospective study enrolled a birth cohort of 411 at-risk children of asthmatic mothers. Spirometry was performed at one month in 403 (98 percent) children and again at age seven in 317 (77 percent).
Significant neonatal airflow deficits, as measured by forced expiratory flow at 50 percent% of vital capacity and forced expiratory volume after 0.5 seconds, were observed among the 14 percent of children who developed asthma by age seven. Bronchial responsiveness to methacholine, which provokes narrowing of the airways, was also significantly associated with the development of asthma. Neonatal airway reactivity was a stronger predictor of asthma than neonatal lung function.
"We found that approximately 40% of the airflow deficit that was associated with asthma in our study was present at birth, while 60% developed through early childhood along with the disease," noted Dr. Bisgaard. "This indicates that both prenatal and early childhood mechanisms are potential intervention targets for the prevention of asthma."
The study used a homogenous study sample, which might limit extrapolation of the results to other populations.
"It seems that lung function changes associated with asthma occur very early in life and maybe even before birth," concluded Dr. Bisgaard. "This may explain the lack of effect from early intervention with inhaled corticosteroids and should direct research into the pathogenesis and prevention of asthma towards the earliest phases of life."

Dynamic Mitochondrial Networks in Cancer

Research projects evolve in a fortuitous manner, often guided by a convergence of novel observations, intuition, helpful colleagues and unique personal circumstances. It is precisely this constellation that prompted two cardiologists to study the mitochondrial networks in lung cancer cells.
In 2008, my colleague and friend Stephen Archer, a Professor of Medicine at the University of Chicago, asked me whether I would be interested in studying the role of mitochondrial networks in lung cancer cells. My first response was the question ?Do mitochondria really form networks??, because at that time the expression ?mitochondria? evoked images of scattered oval-like organelles, a textbook image of electron microscopy.
I was also intrigued by my colleague?s request, since we were both cardiologists and it therefore appeared to be somewhat unusual for us to study cancer cells. However, as is often the case in science research, personal motivations lay behind Stephen?s newfound research interest ? Stephen?s cousin had recently died from lung cancer.
His cousin?s untimely death and Stephen?s frustration at the lack of therapeutic options for lung cancer victims had served as an incentive for him to expand his ongoing work on the role of mitochondria in cardiovascular cells to also include the investigation of mitochondria in cancer cells. Could we contribute to the identification of novel approaches to treat lung cancer?
During the preceding years, Stephen had focused on the role of glucose oxidation in cancer, in part inspired by the work of the German Nobel prize laureate Otto Heinrich Warburg (1883-1970). In the 1920s, Warburg hypothesized that cancer cells primarily rely on non-oxidative glycolysis instead of glucose oxidation to fuel their energy demands.This metabolic signature of cancer cells was critical for the development and growth of tumors.
As he examined the metabolism of malignant lung cancer cells and non-malignant healthy epithelial cells, Stephen had noticed an important difference in the physical appearance of the mitochondria. The mitochondria in the vast majority of cancer cells appeared to be small and fragmented, while healthy epithelial cells predominantly contained elongated, filamentous-like mitochondria that formed large intact networks. The cause and significance of this difference in the mitochondrial structure between lung cancer cells and healthy lung epithelial cells was unknown and thus a fertile ground for new discoveries.
Even though the planned collaborative project would primarily focus on the mitochondrial network structure and not the mitochondrial metabolism of cancer cells, I also decided to read some of the original Warburg papers in the original German language. German used to be a major language of scientific communication and publication in the 19th century as well as the first half of the 20th century. However, during the latter half of the 20th century and especially in the 21st century, English has become the predominant language of the scientific enterprise, even in Germany.
My nostalgic longing for reading scientific articles in German and my curiosity about how scientists wrote articles in the 1920s prompted me to download some of the Warburg papers. I have to admit that I was quite impressed by the comprehensive nature of the work described. The paper entitled ??ber den Stoffwechsel der Carcinomzelle? (Biochemische Zeitschrift 152, 309-344 (1924)) by Warburg and his co-authors Karl Posener and Erwin Negelein contains a comprehensive evaluation of the respiration of tissues from multiple organs, such as the epithelium, connective tissue, brain tissue, retina and various benign and malignant tumors. This 36 page paper includes numerous hypotheses, observations and conclusions about the nature of tumor metabolism that would inspire subsequent generations of scientists.
One observation made by Warburg, for example, toward the end of the manuscript is that tumors with high levels of glycolysis are also associated with high levels of ammonia production and Warburg refers to this observation as an oddity that needs further research. It would take at least 80 years for researchers to understand some of the key underlying molecular mechanisms that explain this observation, when multiple research groups demonstrated that cancer cells use the amino acid glutamine as a major mitochondrial substrate and which upon degradation releases ammonia.
After reading the awe-inspiring Warburg papers, I felt even more enthusiastic about embarking on this new collaboration to study mitochondrial networks in cancer cells.
Peter Toth, a pharmacologist and neuroscientist who directed the confocal microscopy imaging core in Stephen?s group, used his extraordinary live-cell imaging expertise to visualize the mitochondrial networks of malignant and non-malignant lung cells over time. The microscopy data showed that the mitochondrial networks of cells were highly dynamic, continuously undergoing mitochondrial fission (fragmentation or division) and mitochondrial fusion (rejoining). However, at any given time, the majority of cancer cells had smaller, fragmented mitochondria when compared to healthy lung epithelial or vascular cells.
Working with a number of colleagues in our laboratories, we determined that lung cancer cells expressed higher levels of the mitochondrial fission protein Drp-1 when compared to multiple healthy cell types found in the lung. Inhibition of Drp-1 reversed the mitochondrial fragmentation and restored the degree of mitochondrial networking in malignant cancer cells to the levels we observed in healthy non-malignant cells. Similarly, over- expression of the mitochondrial fusion mediator Mitofusin-2 (Mfn-2) also increased mitochondrial networking. Importantly, inhibiting mitochondrial fission resulted in a cell cycle arrest of cancer cells and markedly reduced cancer cell proliferation. In vivo experiments using a tumor xenotransplant model showed a marked reduction in tumor progression when tumors were either treated with a pharmacological inhibitor of Drp-1 or when Mfn-2 was over-expressed.
Our findings are consistent with the observation that mitochondria undergo a cycle of fission and fusion which is coordinated with the cycle of cell division (mitosis). Our experiments suggest that targeting the mitotic fission of mitochondria may be a complementary approach to halt cancer cell proliferation. When we examined the tumor tissues of lung cancer patients, we found that tumor regions indeed expressed markedly higher levels of Drp-1 than healthy lung tissues. Whether Drp-1 levels are also higher in other forms of cancer and whether targeting mitochondrial fission in these other tumor tissues would be equally beneficial still needs to be examined in future studies.
Prior studies have detailed the role of Drp-1 in non-malignant cells where the protein appears to play a role in cell death. Drp-1 induced mitochondrial fission is a characteristic of mitochondrial apoptosis and short-term inhibition of Drp-1 can actually prevent cell death. However, other studies have also linked Drp-1 activation and mitochondrial fission to cell proliferation to mitochondrial fission because the cell cycle regulator Cdk1/Cyclin B regulates the activity of the mitochondrial fission mediator Drp-1. This suggests that mitochondrial fission induced by Drp-1 has two very distinct and nearly contradictory roles: cell death and cell growth.
In the highly proliferative cancer cells that we studied, Drp-1 appeared to be primarily acting as a mediator of mitotic fission, but it is quite possible that in other cell types or settings, Drp-1 may be more closely tied to regulation of apoptotic fission. The fact that the same protein regulates seemingly opposite processes of apoptotic fission during cell death and mitotic fission during cell proliferation may seem surprising. However, it is also reminiscent of the fact that mitochondria themselves can have apparently contradictory roles in cells, acting both as metabolic powerhouses as well as initiators of cell death.
From an evolutionary and teleological standpoint, a cell undergoing division (mitosis) would want to coordinate this process with the dividing and distributing of its mitochondrial organelles. Intact mitochondrial networks are probably difficult to distribute to daughter cells, whereas smaller, fragmented (?fissioned?) mitochondria can be easily distributed. Our study suggests that a reverse signal also exists, by which halting the mitochondrial fission seems to also halt the progression of the cell cycle.
As with any research, our study also points towards many unanswered questions, some of them highlighting the importance of how the nucleus communicates with other organelles: what are the specific mechanisms by which preventing mitotic mitochondrial fission signals back to the nucleus and halts the progression of the cell cycle? How does the cell coordinate the dynamics of other organelles during cell cycle? Could the dynamics of other organelles also be therapeutically targeted in cancer cells?

Sustainability continues to be a hot topic with almost everyone claiming to be some shade of green, which raises the question, ?What does sustainability really mean?? In the world of commercial real estate, sustainability can be broadly categorized into two areas ? construction and operations. ??
1. Construction ? New sustainable construction, tenant fit-outs and major building renovation projects tend to receive the most visibility. In construction related sustainability, the emphasis is on the environmental impact of building the site and interior physical space, as well as the supporting equipment and fixtures. Sustainable construction focuses especially on the use of high-density or urban sites, promoting energy and water efficiency as well as using local and/or renewable materials.
2. Operations ? More recently sustainable operations are receiving greater attention. Rather than focus on environmental quality during construction, companies are looking to ?green? their operations through solutions in transportation and commuting; recycling and waste management; and achieving increased utility efficiency.
However, green construction and operations are just two legs of the sustainability stool ? the third is actual demand for the space, culminating in a lease. Today, tenants have a growing desire (and in some cases, requirement) to be in green spaces. And landlords, especially large public ones, are increasingly looking to be green and share some of the same sustainability concerns as the tenants.? So what drives tenants and landlords to demand sustainability in their real estate leasing programs? By and large it is the triple bottom line: People-Planet-Profit. Tenants need their workforce (People) to be healthy and productive. They want to maximize their bottom line (Profit) and they need a secure and stable place in which to carry out their enterprise (Planet).? A company?s real estate is where the three Ps tend to converge so space use has recently found itself the central focus of corporate green programs.?

How to cook: Sweet Potato Soup with Caramelized Onions
We only serve this sweet, creamy soup throughout the autumn and winter months months. Possibly because of its restricted year, it has created considerably of a cult adhering to. Its personality comes from the caramelized onions floating in each spoonful. After i make this soup, I constantly make further caramelized onions simply because they generate an excellent topping for whipped potatoes at another meal. The onions can cook while you are preparing the rest from the soup. The soup can make a chic initial class for a fancy meal.
Ingredients:
Caramelized Onions
2 tablespoons salted butter
2 large Spanish onions, peeled and thinly sliced
1/2 cup packed dark brown sugar Kosher salt and freshly ground black pepper, to taste
1/4 cup balsamic vinegar
Soup
4 tablespoons salted butter
3 whole cloves garlic, peeled and left whole
2 cups sliced onions
2 cups sliced carrots
1/4 cup diced celery
8 medium-large sweet potatoes, peeled and cut into chunks
10 cups chicken stock
1/2 cup packed brown sugar
1/2 teaspoon ground nutmeg
2 tablespoons Worcestershire sauce
2 cups light cream
1/2 cup sweet sherry wine
Kosher salt and freshly ground black pepper, to taste
11/2 cups caramelized onions
Directions:
For your caramelized onions: In the large saute pan melt the butter more than medium heat. Include the onions and saute for 18 minutes. Include the brown sugar, salt and pepper. Saute an extra 10 minutes. Add the vinegar and proceed cooking until the vinegar commences to evaporate and the onions search thick and syrupy, eight to 10 minutes far more. Remove from the heat and let interesting. If working ahead, the onions could be stored within a tightly lined container till prepared to use.
For your soup: Within a stockpot melt the butter over medium-high warmth. Include the garlic, onions, carrots, and celery. Saute for ten minutes. Add the sweet potatoes and saute 5 minutes a lot more. Pour the inventory above the vegetables and bring to a boil. Reduce the heat to medium and simmer right up until the sweet potatoes are very soft, thirty to 35 minutes. Puree the soup in the pot using a hand blender or operating in batches with a regular blender until clean. Add the brown sugar, nutmeg, Worcestershire sauce, gentle cream,sherry, salt, and pepper. Puree once again till well mixed. Stir the caramelized onions in to the soup with a spoon.
Makes 8 to 10 servings

Huawei Ascend P1 pegged for April release in China
Back during CES 2012 we spent sometime with Huawei's flagship device, the Ascend P1. Shortly after that, there was speculation it would land in China available in late March but it seems that date may have now been pushed to mid-April with other regions getting the device later on. As a reminder, here is what folks will be waiting for:

New physical attraction between ions in quantum plasmas
Nowadays, ever smaller and more powerful computer chips are in demand. RUB physicists have discovered a new physical attraction that accelerates this progress. Prof. Dr. Padma Kant Shukla and Dr. Bengt Eliasson found a previously unknown phenomenon in quantum plasmas. A negatively charged potential makes it possible to combine positively charged particles (ions) in atom-like structures within the plasma. In this way, current can be conducted much more quickly and efficiently than before, opening new perspectives for nanotechnology. The researchers report on their findings in Physical Review Letters.
Electrons and ions in ordinary plasmas
An ordinary plasma is an ionized electrically conducting gas consisting of positive (ions) and negative charge carriers (so-called non-degenerate electrons). This is the chief constituent of our solar system. On Earth, such plasmas among others can be used to produce energy in controlled thermonuclear fusion plasmas similar to the sun, or even to fight disease in the medical application field.
New effect on the atomic scale in quantum plasmas
Quantum plasmas extend the area of application to nano-scales, where quantum-mechanical effects gain significance. This is the case when, in comparison to normal plasmas, the plasma density is very high and the temperature is low. Then the newly discovered potential occurs, which is caused by collective interaction processes of degenerate electrons with the quantum plasma. Such plasmas can be found, for example, in cores of stars with a dwindling nuclear energy supply (white dwarfs), or they can be produced artificially in the laboratory by means of laser irradiation. The new negative potential causes an attractive force between the ions, which then form lattices. They are compressed and the distances between them shortened, so that current can flow through them much faster.
Microchips and semiconductors
The findings of the Bochum scientists open up the possibility of ion-crystallization on the magnitude scale of an atom. They have thus established a new direction of research that is capable of linking various disciplines of physics. Applications include micro-chips for quantum computers, semiconductors, thin metal foils or even metallic nano-structures.

New physical attraction between ions in quantum plasmas
Nowadays, ever smaller and more powerful computer chips are in demand. RUB physicists have discovered a new physical attraction that accelerates this progress. Prof. Dr. Padma Kant Shukla and Dr. Bengt Eliasson found a previously unknown phenomenon in quantum plasmas. A negatively charged potential makes it possible to combine positively charged particles (ions) in atom-like structures within the plasma. In this way, current can be conducted much more quickly and efficiently than before, opening new perspectives for nanotechnology. The researchers report on their findings in Physical Review Letters.
Electrons and ions in ordinary plasmas
An ordinary plasma is an ionized electrically conducting gas consisting of positive (ions) and negative charge carriers (so-called non-degenerate electrons). This is the chief constituent of our solar system. On Earth, such plasmas among others can be used to produce energy in controlled thermonuclear fusion plasmas similar to the sun, or even to fight disease in the medical application field.
New effect on the atomic scale in quantum plasmas
Quantum plasmas extend the area of application to nano-scales, where quantum-mechanical effects gain significance. This is the case when, in comparison to normal plasmas, the plasma density is very high and the temperature is low. Then the newly discovered potential occurs, which is caused by collective interaction processes of degenerate electrons with the quantum plasma. Such plasmas can be found, for example, in cores of stars with a dwindling nuclear energy supply (white dwarfs), or they can be produced artificially in the laboratory by means of laser irradiation. The new negative potential causes an attractive force between the ions, which then form lattices. They are compressed and the distances between them shortened, so that current can flow through them much faster.
Microchips and semiconductors
The findings of the Bochum scientists open up the possibility of ion-crystallization on the magnitude scale of an atom. They have thus established a new direction of research that is capable of linking various disciplines of physics. Applications include micro-chips for quantum computers, semiconductors, thin metal foils or even metallic nano-structures.

Treating Arthritis With a Good Arthritis Supplement
A good arthritis supplement can help alleviate the painful and debilitating symptoms of arthritis. Arthritis is a condition that commonly affects the joints and muscles. The most common symptoms of arthritis are stiff and painful muscles and joints.
What Happens When You Have Arthritis?

It appears that results from using such a supplement differ from one person to the next. If you suffer from arthritis, then there is no harm in trying such supplements. If, after three months, you do not feel any positive effects, then you should discontinue it.
Methyl sulfonylmethane (MSM) is another supplement that is gaining in popularity. It is not as well established as glucosamine and chondroitin, however. There have been fewer scientific studies done regarding the effects and efficacy of MSM supplements. You should therefore be very careful about using such supplements.
Side Effects of Arthritis Supplements
While most people encounter no side effects from glucosamine and chondroitin, a small percentage do. These side effects include nausea, flatulence, diarrhoea, constipation, and heartburn. If you experience any of these side effects when you start taking any kind of arthritis supplement, you should stop immediately.
Some Things To Take Note Of
If you also suffer from diabetes, then you should be very careful when taking any type of arthritis supplement. This is because the supplements also raise your blood sugar levels. People suffering from diabetes must control their blood sugar levels carefully. If you think that your body cannot handle so much extra sugar, then you should not take arthritis supplements.
Also, if you have to take anticoagulant medication, then you should not take any supplement. The effects of mixing these two drugs may have a harmful effect on your body.
All in all, a good supplement can be of great help to you in fending off the symptoms of arthritis. Those people for whom these supplements work find that they are able to function better because of them. When you go shopping for a good arthritis supplement, make sure you buy them from a reputable company.
